the three big problems of natural science

It is said that the three big problems of natural science are these three questions listed below.
1,genesis of the universe from the "null"
2,appearance of organic matters from minerals
3,sprout of consciousness to life

The second one can be explained with the fact that RNA is a kind of von-neumann machine. By contrast, the first question is only partly solved,because the other part of this issue is related to the dark matter/energy problem, and superstring theory. The third one had not been recognized as the problem of natural science in 20th century, however recent development of neuroscience made it as a field of science. I hope some kind of theory of this question will appear soon.
